There was a time when to amplify DNA,
You had to grow tons and tons of tiny cells.
Then along came a guy named Dr. Kary Mullis*,
Said you can amplify in vitro just as well.
Just mix your template with a buffer and some primers,
Nucleotides and polymerases, too.
Denaturing, annealing, and extending.
Well it's amazing what heating and cooling and heating will do.
PCR, when you need to detect mutations.
PCR, when you need to recombine.
PCR, when you need to find out who the daddy is.
PCR, when you need to solve a crime.
*Kary Mullis，1993 年因發(fā)明了PCR技術(shù)獲得諾貝爾化學獎。
要翻譯過來，大概就是這么說的：
以前如果要擴增DNA，就得培養(yǎng)很多很多的那小小的細胞（細胞帶有目的DNA，屬于體內(nèi)擴增)。
后來有個叫Kary Mullis 的家伙跑出來說，其實你可以在體外很簡單的達到目的。
只要把你的模板DNA 跟緩沖液混勻了，當然還要加一些引物，核苷和聚合酶。
然后，變性（94 度，讓DNA 雙鏈變性成單鏈）、退火（55 度左右，讓引物可以跟匹配的DNA 序列結(jié)合）、延伸（72 度，聚合酶的工作溫度，目的DNA 片斷被復(fù)制），你會發(fā)現(xiàn)加加熱降降溫也有奇妙的效果。
當你想檢測突變時，來做PCR 吧；
當你想重組基因時，來做PCR 吧；
當你要親子鑒定時，來做PCR 吧；
當你要解決案件時，來做PCR 吧。
